Harley as I see her is a very straightforward rogue character. She is very charismatic, and athletic, so high Charisma and Dexterity are obviously a must. In the comics, she is given a potion by Poison Ivy to also make her abnormally strong and resilient to damage, so decent strength and constitution also work. Intelligence and Wisdom are not her strong suits, although she is a doctor, so her intelligence shouldnt be a negative. Her wisdom score is likely to be her lowest score, as she acts very insane and does somewhat unorthodox things.
Skills she'd have would be Bluff, Diplomacy, Intimidate, Balance, Jump, Tumble, Escape Artist, Use Rope, Perform, Picket Pocket, Hide, Move Silently, among others.
She carries around a super large hammer too, so a feat into Exotic Weapon Proficiency: Maul would give you that. She also has a pop-gun pistol, but you may not have guns in your campaign, so substitute that with a hand-crossbow, and the feats that let her fire into combat without penalty.
She basically uses her rogue sneak attacks whenever possible, and I've repeatedly seen her whack Batman with that monstrous hammer in the back of the head, knocking him out or silly.
Use her tumble skills to get her into position for those sneak attacks.
Feats I'd recommend are Improved Initiative, Exotic Weapon: Maul, Combat Reflexes, Lightning Reflexes, Point Blank Shot, Precise Shot, Rapid Reload.

You could really have fun with a character like this in almost any DnD setting.

Also, if you really wanted to make her more of a charmer character, add some ranks into Use Magical Device, or add a few levels into Bard, so you can use the cool spells they get, like Charm Person to stop that nasty Batman from whoopin on you and Mistah J. :D
