If a Duskblade is wielding a 2 handed weapon and wishes to cast a spell, can he take one hand off the weapon, cast the spell and return his off hand to the weapon to be able to threated with the weapon?

Yes, I believe he could, however he would not threaten while casting the spell. This would only rarely be a problem, such as if someone had readied an action or was close enough to get an AoO off of you for casting and they did something which normally provokes an AoO (grapple, bullrush, etc) then you wouldn't get to take that AoO.

It is a free action to switch grips, which is what you are describing.
