what effect do bucklers and two-handed weapons have on eachother? does using a two handed wep negate any bonus to ac the bucklet gives?

You get no armor bonus if you've used a 2h weapon unless you take the feat. "Improved buckler defense"?

That is the feat, and it doesn't remove the -1 penalty to attack rolls (which is not armor check). Don't get the feat until you can afford a +1 buckler to compensate for the loss of attack.

Just get a mithral buckler and be done with it. No more -1 check penalty. :P

You still need the feat to get both the Buckler AC bonus and the use of the hand to attack.

Even a mithral buckler applies a -1 penalty to attack rolls for 2H or off-hand weapons.

Normal THW with Buckler: No AC, -1 attack for using buckler hand, -ACP attack
With Improved Buckler Defense: -1 attack for using buckler hand, -ACP attack

Note that it is very easy to get a Buckler without any ACP.

You only take the ACP to attacks if you aren't proficient with bucklers, of course.
