Is it reasonable to remove the standard death attack of an assassin and grant the assassin the following special attack to make the class a better martial one:

1) 1/round (instead of one normal attack) if the attack hits the victim must make a successful fortitude save DC 10 + assassin level or suffer 1d6 (per 3 assassin levels) constitution damage

2) above but instead of a full round attack

3) as 1) but only after a successful sneak attack

4) as 1) but only in case of a critical hit

I wouldn't accept it... it negates the whole concept of death by one strike

Which is really the only point of taking levels in Assassin.

(Okay. I'm sure there are other good things as well, I just can't think of them.)

Yeah, assassins who find themselves in a melee fight have already failed.

The ability to cast Arcane spells without preparation using Int while wearing (light) armor (and with faster spell level progression than a bard)? Less useful when splatbooks are allowed, but even then it's nice for many builds, especially since (1) it's relatively easy to qualify for and (2) it grants Sneak Attack, unlike Beguiler and Duskblade.

If nothing else, I'd use it for Dragon Disciple builds on occaison.

I usually make my assassins monks. Stunning Fist goes nicely with sneak attack. Throwing a flurry of poisoned shurikens while invisible is pretty neat too.
