That... sounds freaking awesome. I've loved Joss' work for a while now, and he's great at the whole "strong female characters" thing. If he had time to plan something, it would probably twist a few adventure stereotypes and could be hilariously fun.

I've got a few people in mind (J. R. R. Tolkien could make a nice DM), but my preference would be to give Mary Shelly a turn behind the screen. I'd also like to see what some of my favorite anime/manga authors could do at the table.

Seriously though, they got a raw deal (both shot and killed) then, and under really messed up circumstances, so I'd like to see them get a fair break later. I posed this question to the guys I work with and game with. While they chose to flesh out an entire party of famous & historical people, rather than just naming one person, I thought I'd share their suggestions:

Party One:

DM -- Alexander the Great

Spartacus as the slave turned fighter
Hugh Laurie as the sarcastic rogue
Sir Alec Guinness as the wizard/veteran of the last war
Jesus Christ as the cleric/party heal-monkey
Bruce Lee as the butt-kicking monk
Axl Rose (or Steven Tyler) as the strung-out bard
Megan Fox as the archery ranger and token hot chick


Party Two:

DM - Noah "Spoony" Antweiler
Brad Pitt as a halfling rogue
Stephen Fry as a wizard

Party Three:

DM : George R R Martin.

Fighter (or Paladin) - Winston Churchill
Druid - David Suzuki
Wizard - Cao Cao (Chinese warlord from the Romance of the Three Kingdoms)

Genghis Khan as a barbarian/brava warlord
Aristophanes as a halfling rogue
Homer as the DM
John Paul the second as a laser cleric
Rasputin as a warlock
and me as whatever the party needs
now if only all of them could work together it would be great.

Given that I'd much rather have people who I'm confident would enjoy it (and actually play, or I suspect might play/want to play), I'd go with Vin Diesel, Julia Ling, Stephen Colbert, Felicia Day, David Boreanaz, Bruce Campbell, and/or Lauren Graham.

I'm pretty sure that if David Boreanaz were at the table with me, I'd be too busy trying to make him fall desperately in love with me to actually pay attention to the game.

Oddly enough, as much as I love Johnny Depp, I find his gorgeousness less intimidatingly overwhelming. It'd be like, "Johnny, you're hot. Leave that Vanessa chick and come be with me? No? Ok, well, can't blame a girl for trying, can you? Let's play some D&D."
