With Giant killer, is your reaction all your attacks?  I have 2 attacks. Does that mean I attack twice for a reaction and again twice when it's my turn?

 

With Multiattack Defense, if 2 or more creatures hit me, do you gain +4 AC to all the attackers?

 

For Whirlwind Attack, does that apply to only opponents next to me or can I use my movement and keep attacking creatures on the way

The reaction granted by Giant Killer is a single attack - your normal ability to attack multiple times is tied to the Attack action, which is a specific action that you take on your turn.

 

If multiple creatures with multiple attacks are attacking you, you can gain the AC bonus against each of them individually - you would not apply the +4 bonus to AC against the second creature's attack unless it had hit you with an attack, regardless of whether the first creature had hit you or not.

 

You can move to position yourself within 5 feet of the largest possible number of monsters, but I do not believe that you could use whirlwind attack to attack the creatures within 5 feet of you at point A and then move to point B and attack the creature within 5 feet of you at that point.

For Colossus Slayer and Horde Breaker, they both specify either add damage once or gain an extra attack once.

For Giant Slayer, it doesn't state any limitation. It's states that you attack as a  reaction. I know you are allowed one action in general. However, there are exceptions to rules based on characters. I.E. The sorcerer using its Quickened ability to cast 2 spells on one turn.

Nov 20, 2014 5:44:38 		Your example doesn't count since that was already possible (Action Surges, bonus action spells). Also Sorcerers use their bonus action for the quickened spell, just like this uses your reaction. In both cases you can only use it once because you only have one bonus action/reaction.
